Eloralintide

By Hack SmithLast updated: How we research

Obesity and weight management, with specific interest in dual/triple combination with GLP-1 and GIP agonists

Eloralintide (LY3841136) is a long-acting amylin receptor agonist in clinical development by Eli Lilly. Amylin is a pancreatic peptide that works alongside insulin to regulate postprandial glucose and satiety. Eloralintide activates amylin/calcitonin receptors to slow gastric emptying, promote satiety, and reduce food intake through a mechanism complementary and distinct from GLP-1 agonists. Early-phase clinical data suggest meaningful weight loss, particularly when combined with GLP-1 agonists such as tirzepatide or semaglutide. Combination studies are ongoing as the complementary mechanisms may provide additive efficacy with manageable GI side effects.

Common Uses
  • Weight Management
  • Appetite Suppression
Evidence Grade

Early-phase human data (Lilly); limited public data.
Safety Notes

Emerging compound, limited human safety data; GI effects expected for the amylin class.

Mechanism of Action

A long-acting amylin receptor agonist (amylin analog) in clinical development by Eli Lilly. Activates amylin/calcitonin receptors to slow gastric emptying, increase satiety, and reduce food intake — complementary to, and distinct from, GLP-1 agonists, so it's studied both alone and stacked with incretin agents for weight management.
