- Depression/Mood Support
- Anxiety Reduction
- Neurogenesis
No human clinical trials have been conducted - safety profile in humans is completely unknown This is an experimental research compound not approved for human use TREK-1 channels are expressed in multiple organ systems including heart and nervous system - systemic effects are unpredictable
Mechanism of Action
PE-22-28 is a shortened synthetic analog of spadin that potently inhibits the TREK-1 potassium channel with an IC50 of 0.12 nM. TREK-1 is a two-pore domain potassium channel that, when blocked, increases neuronal excitability and appears to enhance monoaminergic neurotransmission in brain regions associated with mood regulation. Key Pathways: TREK-1 potassium channel inhibition, Enhanced neurogenesis (new neuron formation), Increased synaptogenesis (new synaptic connections), Monoaminergic neurotransmission modulation By blocking TREK-1 channels, PE-22-28 may produce rapid antidepressant-like effects through a mechanism distinct from traditional SSRIs. The compound's ability to promote neurogenesis and synaptogenesis in animal models suggests potential for structural brain changes that could support sustained mood improvement, though this remains unproven in humans.
