Mechanism of Action

P-21 is a small synthetic peptide derived from a small active fragment of ciliary neurotrophic factor (CNTF) — not from Cerebrolysin. It is based on the neurotrophic region of CNTF and increases BDNF production and promotes neurogenesis. P-21 also inhibits beta-amyloid aggregation through interactions with amyloid precursor protein pathways. Key Pathways: CNTF receptor axis signaling (BDNF upregulation), Neurogenesis promotion in hippocampal regions, Beta-amyloid aggregation inhibition Animal studies have demonstrated significant cognitive improvements in models of aging and Alzheimer's disease, with effects on learning and memory that persist after discontinuation, suggesting synaptic remodeling rather than acute receptor occupation. Unlike direct CNTF administration, P-21's small fragment size allows CNS penetration without the systemic side effects of full-length CNTF.
