FOXO4-DRI
Clears aging cells for tissue renewal
- Anti-Aging/Longevity
- Tissue Regeneration
No human clinical trials have been conducted - all safety data is from animal/in vitro studies Animal research shows senescent cell elimination can worsen pulmonary hypertension and increase vascular remodeling Senescent cells serve beneficial functions in some tissues - indiscriminate elimination may cause harm This is an experimental research compound, not approved for human use in any jurisdiction
Mechanism of Action
FOXO4-DRI is a synthetic D-retro-inverso peptide that disrupts the interaction between FOXO4 and p53 proteins specifically in senescent cells. In senescent cells, FOXO4 sequesters p53 in the nucleus, preventing p53 from triggering apoptosis; FOXO4-DRI competitively binds to p53's disordered transactivation domain, releasing p53 to restore its normal pro-apoptotic function and selectively induce senescent cell death. Key Pathways: FOXO4-p53 protein-protein interaction disruption, p53-mediated apoptosis activation in senescent cells, Reduction of senescence-associated secretory phenotype (SASP) By selectively eliminating senescent cells while sparing healthy cells, FOXO4-DRI aims to reduce the chronic inflammatory burden and tissue dysfunction caused by accumulated senescent cells during aging. In animal studies, this has translated to improved tissue homeostasis, restored testosterone production in aged testes, improved spermatogenesis, and reduced pulmonary fibrosis.
