Mechanism of Action
MOTS-c is a 16-amino acid peptide encoded by mitochondrial DNA that functions primarily through activation of the AMPK (AMP-activated protein kinase) pathway, a master regulator of cellular energy homeostasis. It enhances mitochondrial biogenesis and function, modulates oxidative stress responses by increasing antioxidant enzymes (SOD, CAT), and inhibits inflammatory signaling cascades including the MAPK pathway (ERK, JNK, p38 phosphorylation). Key Pathways: AMPK activation and energy metabolism regulation, MAPK signaling pathway inhibition, Mitochondrial biogenesis and MAVS signaling, Antioxidant defense systems (SOD, CAT, GSH restoration), PPAR pathway modulation affecting lipid metabolism Through AMPK activation, MOTS-c improves glucose and lipid metabolism, which may explain observed associations with better metabolic profiles and reduced fatty liver markers. Its effects on mitochondrial function and oxidative stress reduction connect to the observed protection against tissue damage in various organs and potential preservation of muscle mass during disuse.
