Hexarelin
Boosts growth hormone for better sleep
- Muscle Growth/Strength
- Fat Loss
- Sleep Improvement
May elevate cortisol and prolactin levels - monitor if using long-term Strong appetite stimulation expected due to ghrelin receptor activation Avoid concurrent NSAID use - may worsen NSAID-induced gastric injury Human safety data is limited - most evidence comes from animal studies
Mechanism of Action
Hexarelin is a synthetic growth hormone-releasing peptide (GHRP) that binds to and activates the growth hormone secretagogue receptor (GHS-R1a), stimulating growth hormone release from the pituitary gland via a Gq-coupled signaling pathway that increases intracellular calcium. Additionally, hexarelin binds to the CD36 scavenger receptor, which mediates distinct cardiovascular and metabolic effects independent of GH release, including activation of PPARgamma pathways that regulate cholesterol metabolism. Key Pathways: GHS-R1a receptor activation → Gq signaling → increased intracellular calcium → GH release, CD36 scavenger receptor binding → PPARgamma activation → cholesterol efflux regulation, Akt/GSK-3β phosphorylation pathway → anti-apoptotic/neuroprotective effects The GHS-R1a activation drives growth hormone release, which theoretically supports muscle growth and fat metabolism. The cardioprotective effects observed in heart failure models appear mediated through both GH-dependent and GH-independent pathways, including reduced cardiomyocyte apoptosis and improved ventricular function. Neuroprotective effects seen in animal models involve Akt pathway activation and reduced caspase-3 activity.
