Safety Notes

No long-term human safety trials exist — use represents self-experimentation with unknown chronic risks Individuals with diabetes, pre-diabetes, or insulin resistance should exercise caution due to potential effects on glucose metabolism Those with history of cancer or active malignancy should avoid due to theoretical concerns about GH/IGF-1 and tumor growth Quality control is a significant concern — peptide purity and authenticity cannot be verified in unregulated markets

Mechanism of Action

MOD-GRF (1-29), also known as CJC-1295 without DAC (Drug Affinity Complex), is a synthetic analog of growth hormone-releasing hormone (GHRH) that binds to and activates the GHRH receptor on pituitary somatotroph cells. This binding stimulates the release of endogenous growth hormone (GH) in a pulsatile manner, mimicking natural physiological secretion patterns. The 'modified' designation refers to amino acid substitutions at positions 2, 8, 15, and 27 that increase resistance to enzymatic degradation by dipeptidyl peptidase-IV (DPP-IV), extending its half-life from minutes to approximately 30 minutes. Key Pathways: GHRH receptor activation on anterior pituitary somatotrophs, cAMP/PKA signaling cascade leading to GH gene transcription and secretion, Downstream IGF-1 axis activation in liver and peripheral tissues, Hypothalamic-pituitary GH feedback regulation By stimulating pulsatile GH release, MOD-GRF (1-29) is theorized to enhance protein synthesis, lipolysis, and tissue repair processes associated with growth hormone activity. Users report improved sleep quality, potentially due to GH's role in slow-wave sleep regulation, along with gradual improvements in body composition. The pulsatile release pattern is considered more physiological than exogenous GH administration, potentially reducing negative feedback suppression.
