Peptide Cycling 101: On/Off Cycles, Duration, and Why It Matters

By Hack SmithLast updated: Not recordedHow we research

When and why to cycle peptides — receptor desensitization explained, cycling protocols for GH secretagogues, BPC-157, Epitalon, MK-677, and more.

Cycling — structured periods of use followed by deliberate off-periods — is one of the most important and most misunderstood aspects of peptide protocols. It's not just about "taking breaks." The rationale differs completely between compound classes: GH secretagogues desensitize their receptors with continuous stimulation; some peptides are inherently course-based by design; others require no cycling at all. Getting this wrong means either a protocol that quietly stops working, or unnecessary breaks from compounds that don't need them.

Why Cycling Matters: The Receptor Desensitization Problem

GHRH and ghrelin receptors respond to repeated stimulation by reducing their sensitivity — a process called receptor downregulation or desensitization. For GH secretagogues like Ipamorelin and MOD-GRF, continuous daily use for 16+ weeks leads to progressively diminishing GH pulse responses. The off-cycle period allows receptor expression to recover, so the next on-cycle starts with full sensitivity again.

Not all peptides work through receptors that desensitize this way. BPC-157 works through growth factor upregulation and angiogenesis — mechanisms that don't have the same desensitization pattern. GLP-1 agonists are managed by physicians for continuous use. The correct cycling approach depends on the mechanism, not a blanket rule.

Planning Multiple Compound Cycles

When running multiple compounds simultaneously, cycle them together rather than staggering off-cycles independently. For example: Ipamorelin + MOD-GRF stack → 12 weeks on, 4–6 weeks off as a unit. Running one compound through another's off-period adds complexity without clear benefit and makes it harder to assess which compound is driving results.

Exception: compounds with different purposes can run independently. You might cycle GH secretagogues while running BPC-157 continuously for an active injury, since they work through entirely different systems with no desensitization crossover.

Frequently Asked Questions

How do I know if a compound has desensitized — is there a symptom?

The most common report is progressively diminishing sleep quality and recovery improvements from GH secretagogues — the same dose that produced noticeable effects in week 2 produces no subjective effect by week 14–16. Objectively, serum IGF-1 measurements can detect declining GH output over time. Most practitioners cycle on schedule rather than waiting for subjective desensitization because preventing the drop is easier than recovering from it.

Will I lose my progress during the off-cycle?

Progress from GH secretagogues (body composition improvements, sleep architecture changes, connective tissue health) is largely retained during a 4–8 week off-cycle, provided training and nutrition remain consistent. The off-cycle is also when you can evaluate your baseline — many people report their sleep and recovery remain better than pre-protocol even during the break, which confirms the on-cycle produced lasting improvements.

Does BPC-157 really not need to be cycled?

BPC-157 works through receptor-independent mechanisms (growth factor upregulation, NO pathway, angiogenesis) that don't follow the GHRP desensitization model. The cycling recommendation for BPC-157 is based on healing timelines — you use it for the duration of the injury recovery, then stop because the healing process is complete. Some practitioners use it continuously at lower preventive doses (100–200 mcg/day); current evidence doesn't suggest desensitization concerns for this approach.

Is it safe to start a new cycle immediately after an off-cycle?

Yes — the off-cycle is specifically designed to restore receptor sensitivity, and starting the next on-cycle after the off-period is the intended protocol. Many experienced users run 3 cycles per year (12 weeks on / 4 weeks off × 3), or 2 longer cycles (16 weeks on / 8 weeks off × 2).

Do I need to taper off GH secretagogues at the end of a cycle?

No tapering is required for GH secretagogues — unlike GLP-1 agonists or corticosteroids, there's no rebound effect or hormonal dependency from stopping GH secretagogues. You can stop at the end of the cycle without transitioning off. Some people reduce frequency in the final week as a personal preference, but it's not pharmacologically necessary.
